Book excerpt: Stable isotopes (2H, 13C) are useful for studies on the metabolism of endogenous or exogenous compounds. GC/MS makes it possible to measure partial loss of 2H by exchange from polydeute-rated molecules and to detect compartmentation of precursors. Oxidoreduction can be studied by measurements of the loss of isotope from the carbon carrying the hydroxyl group. Label in the coenzyme can be corrected for by the use of mixtures of differently labelled compounds. Isotope effects can be used to study mechanisms in vitro and pathways in vivo. Labelling in vivo with 2H-substrates makes it possible to distinguish coenzyme pools.
